Probably, the best tweaking for the beginner after your system is up and running includes:
1) a full disk scan and defragment of all partitions (Start, Programs, Accessories, System Tools, Defrag)
2) disk clean-up (Start, Programs, Accessories, System Tools, Disk Clean-up)
3) a registry cleaner (found in such programs as Norton's Systemworks,
4) a full system virus scan after you have updated virus definitions
5) a full spyware and adware scan by such free programs as SpyBot and Ad-Aware.
These tweaks should be employed often (weekly prehaps) to keep your system clean and clutter free. And, it goes without saying that you should always check for updates on your operating system's update site, and your anti-virus program's virus definitions update site.
